Is it possible to become a truck driver if I had a DUI 2 years ago?
It was my first and only. I have no other record besides that.

It doesn’t hurt to try.
You need to mention it though in your application if it is asked of you. They WILL find out and it is better they hear your story, then assume you were trying to hide it from them.

you can get a cdl provided you have met all the courts obligations with regard to your dui. However,many companies will not touch you till at least 5yrs has passed,and your driving record impeccable. Alot of companies won’t even touch you after that,because you are considered at risk. There are smaller companies that would hire you after a dui provided you have proven safe driving for sometime after the incident
